ABSTRACT

<p><b>PURPOSE</b>RNA helicase p68 plays an important role in organ development and maturation through tuning cell proliferation. However, the character and role of p68 in the whole wound healing process need more study.</p><p><b>METHODS</b>First, we characterize expression of p68 in normal rat skin development postnatal. Then, we assayed dynamic change of p68 in rat skin from different stage after injury, and explored the role of p68 in proliferation and migration of three types of wound healing related cells.</p><p><b>RESULTS</b>p68 was down-regulated during skin developmental and maturation process, up-regulated after wound, peaked on day 14 and then significantly decreased. Wound fluid enhanced wound healing related cell proliferation and up-regulated expression of p68. Conversely, reducing p68 expression by RNA interference resulted in significantly slower proliferation and migration.</p><p><b>CONCLUSION</b>Our results define an important role of RNA helicase p68 in skin wound healing process.</p>

ABSTRACT

Objective To provide theoretical reference for the research on lateral postural control in older people by using the electromyography (EMG)technique to compare the neuromuscular reaction between the young and older people during lateral perturbation. Methods Postural reactions of 14 young people and 14 older people as subjects were evoked by sudden unpredictable medio lateral translation platform. The surface EMG data were collected from peroneus longus, anterior tibialis, gluteus medius and erector spinae of the left side of the body. Results The latencies in anterior tibialis, gluteus medius and erector spinae were obviously delayed in older people, and the duration of time to peak in peroneus longus, anterior tibialis and gluteus medius of older people was much longer than that of young people. Conclusions The delayed response of ankle, hip and trunk muscle and the low efficacy of contraction rate in ankle and hip muscles to the unexpected perturbation might be the primary reason for the decrease of lateral postural control in older people.

ABSTRACT

<p><b>BACKGROUND</b>Maintenance of normal cardiac function is controlled by the autonomic nervous system. In congestive heart failure (CHF), sympathetic nerve denervation is increasingly recognized. The sympathetic fiber density depends on the balance between neurotrophins and neural guidance molecules. Semaphorin 3A (sema3a), a secreted neural guidance factor, is a well characterized member of the newly found semaphorin family. It can induce sympathetic growth cone collapse and axon repulsion. We conducted this study to investigate cell sources of sema3a in the heart, the expression level of sema3a in CHF and discuss the possible role of sema3a in CHF.</p><p><b>METHODS</b>Rats were divided into four groups: 30 days control group rats, 30 days CHF rats, 60 days control group rats, 60 days CHF rats. The heart failure model was induced by injection of isoproterenol (ISO) 340 mg/kg continuously two days. All animals underwent echocardiography and haemodynamics measurements. Cardiac expression of sema3a was determined by real time polymerase chain reaction (RT-PCR) and Western blotting analysis. Immunohistochemical analysis was used to determine the cell source of sema3a in the heart.</p><p><b>RESULTS</b>Isoproterenol induced 30 days and 60 days CHF rats displayed left ventricular dilation, systolic and diastolic function decrease. Sema3a was secreted by the cardiocytes and increased significantly in 30 days and 60 days CHF rats compared with the controls (RT-PCR: 30 days group: 0.32 ± 0.05 vs. 0.58 ± 0.06, P < 0.01; 60 days group: 0.34 ± 0.08 vs. 0.71 ± 0.07, P < 0.01. Western blotting: 30 days group: 0.25 ± 0.10 vs. 0.46 ± 0.10, P < 0.05; 60 days group: 0.29 ± 0.10 vs. 0.55 ± 0.16, P < 0.01. Immunohistochemical analysis: 30 days group: 2.91 ± 0.20 vs. 5.31 ± 0.30, P < 0.01; 60 days group: 2.94 ± 0.30 vs. 5.80 ± 0.30, P < 0.01).</p><p><b>CONCLUSIONS</b>Sema3a was expressed in the heart by cardiocytes. Increased expression of sema3a may partly account for sympathetic denervation in CHF; modulation of this pathway may prove beneficial in heart failure sympathetic remodeling.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the specific regularity of acupoints to treat genital system diseases.</p><p><b>METHODS</b>A model of inflammatory ovary was developed by injection of 0.08 mL, 10% mustard oil into the ovary of the rat. And the reactive points of plasma extravasarion points on the skin were detected after injection of 0.25 mL evens blue into the tail vein. The relation among the regions of plasma extravasation points, dorsal ganglion of governing ovaries and the acupoints to treat the ovarian diseases was analyzed.</p><p><b>RESULTS</b>The exudative points mainly distributed at Guanyuan (CV 4)-"Zigong" (EX-CA 1) point-area, "Shenshu" (BL 23)-"Mingmen" (GV 4) point-area, "Changqiang" (GV 1)-"Huiyin" (CV 1) point-area, "Sanyinjiao" (SF 6)-"Zusanli" (ST 36) point-area, "Zhongwan" (CV 12) point-area, "Gansho" (BL 18) area, with the most in the "Guanyuan" (CV 4)-"Zigong" (EX-CA 1) point-area and next at "Shenshu" (BL 23)-"Mingmen" (GV 4) point-area.</p><p><b>CONCLUSION</b>The reactive point-area of plasma extravasation resulted from inflammation of ovary are identical to the ovary a corr.sponding dorsal ganglion. Acupuncture at the corresponding acupoints of the reactive point-areas has specific regulative action on ovarian functions.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the specific regulative law of electroacupuncture (EA) for the hypothalamus-pituitary-gonad axis.</p><p><b>METHODS</b>The adult female SD rats were divided into a simulant EA group, Guanyuan (CV 4) group and Neiguan (PC 6) group according to 4 stages of the estrous cycle. Acupuncture was given from 9 : 30 AM to 10 : 00 AM and the brain tissue samples were collected at 15 : 00 PM. GnRH expressions in the nuclei related with genital neuroendocrine in the hypothalamus were investigated with immunohistochemical method.</p><p><b>RESULTS</b>(1) The GnRH expression quantity was different in different estrous cycles in the simulant EA group, with more expression in the proestrus and the estruation; (2) After acupuncture, the GnRH expressions in the medial preoptic area, the arcuate nuclei and the nuclei periventriculares were increased in different stages of the estrous cycle in varying degrees, with the "strong" GnRH expression increased significantly in the diestrus and the metaoestrus in the Guanyuan (CV 4) group.</p><p><b>CONCLUSION</b>After EA, the expression of the GnRH-positive reactant in the medial preoptic area, the arcuate nuclei and the nuclei periventriculares of the hypothalamus significantly increases, with the Guanyuan (CV 4) group better than the Neiguan (PC 6) group, and more obvious expressions in the diestrus and the metaoestrus.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a method for HPLC fingerprint determination of different growing areas on Fructus Aurantii fried with bran.</p><p><b>METHOD</b>HPLC chromatographic experiment was carried out by ultrasonic distilment of acetacetic ester, gradient elution mode and the detection wavelength of 320 nm. The column temperature was set at 30 degrees C, and flow rate is 1.0 mL x min(-1). Naringin served as the reference standard. Ten batches of Fructus Aurantii fried with bran in the different places were analyzed. Similarity evaluation system for chromatographic fingerprint of TCM proceeded the evaluation, The correlation coefficients between ten batches were used to define the similarity.</p><p><b>RESULT</b>There were eleven characteristic peaks in Fructus Aurantii fried with bran by HPLC fingerprint which constituted fingerprint characters. Both the reproducibility and stability on HPLC fingerprint were proper.</p><p><b>CONCLUSION</b>The method was accurate and simple. It can be applied to the analysis on Fructus Aurantii fried with bran. The fingerprint spectrum can be used to distinguish Fructus Aurantii fried with bran and to be its quality control.</p>
